Apple polyphenol was selected as an objective of study, and extraction technology was optimizated, the crude apple polyphenol was separated and purificated by macroporous resin and the anti-radiation function of the apple polyphenol was studied further in this research.Using ultrasound-assisted extraction method, apple polyphenols of Gala was extracted, and using response surface analysis extraction process was optimized, at the end the best extraction process of apple polyphenol was obtained: ultrasonic time 37.51 min, the temperature 41.18¡æ, acetone concentration of 69.36 percent, the ratio of 1:5.21. In this condition, theoretical yeild of the apple polyphenol was up to 1.170 mg/g. After ultrasound-assisted extraction and ethanol purify settlement, apple polyphenol solution was still crude, and apple polyphenol powder with the taste of apple could be obtained by macroporous resin AB-8. FC determination of the purity was up to 79.5 percent, the yield up to 65.1 percent. There were not n-hexane, benzene, toluene and xylene, o-xylene, styrene and other organic solvent residue in the purification of apple polyphenol by the headspace gas chromatograph.Respectively using DPPH method, methyl-purple method, auto-oxidation of pyrogall method, DPPH radical¡¢O2- and ?OH were scavenged by the purification of apple polyphenol. The experimental results showed that: DPPH radical, O2- and OH were significantly scavenged by the purification of apple polyphenol, and the IC50 value was 0.0034 mg/mL, 2.86mg/mL, 0.75mg/mL.Constructed radiation model and determined 5 Gy¦Ã-ray as the best dose of the immune system radiation damage. Compared with the control group, added the purification of apple polyphenols before irradiation, the carbon clearance index¡¢the serum hemolysin¡¢the swelling degree of the paw¡¢and the index of spleen had increased, 1000 mg/kg/d was significantly in statistics. And the content of MDA and the activity of SOD and GSH-Px in the liver of irrated mice was markedly improved, and 1000 mg/kg/d was statistical significance.
